The motherboard is built in wifi. I need to do something before install tplink?

Disable m/b network features within m/b BIOS, if I knew which m/b you have, I could advise on how to access the BIOS where what needs disabling. Some m/b will automatically disable once it detects the new hardware but it depends on the system....

Will this work with a large external antenna on only ONE of the screw on inputs.

Not properly LTE needs two antennas.

Dows it work on windows 10 though it states that its only compatible up til windows 8

Is this strong enough to maintain a signal on the second floor?

Yes, my PC is downstairs and the wi-fi router is upstairs and it works perfectly well. It connects as soon as the PC is switched on.
