Are these gems made of glass

No. It seems like plastic or something

I dont understand how to use, i poke the gems with either end and nothing happens ???

The tip is WAX you need to press gently but firmly to pick up the gems

Can you use these with a hotfix tool please

We have, but its difficult to get such a small amount of glue as these gems are quite small.
