If you already have the board, and a 6th generation processor, you'll need to flash to the most recent BIOS before installing the i3-7100. However, if you already have a 6th generation chip, their isn't much point in buying the i3 7100. If you don't have a board, find a Z270, H270, or B250 board instead. It will work without flashing the BIOS.

Should I get the core i3-7100 or the Pentium G4560 (for the current prices) ?

Higher processor speed and lower wattage made the i3 my choice over the G4560.

Yes but you will have to boot it with a 6th gen skylake processor first, then upgrade the BIOS to F20, before it will work with that processor.
