Q: Can I use a sharpie on these and not have the lettering smudge?

The material is almost in a laminate covering, which make it smudge proof when printed on with a laserjet or similar printer. A sharpie may write on it, but I’m not sure how it would wear beyond innitial use. I had the same thought, but found it was better to print directly to them

Q: Can you print with inkjet

No, you need a laser printer. I was led to believe you could use ink jet. Could not even use a pen, ink never dries and smears.

Q: is there a software or template needed to print on this? where do you get template?

I bought the product a couple years ago. It comes with a link that allows the user to print the labels. Problem was, the inner and outer-most groups print outside the intended labels. I concluded that the best use of the labels would entail the user to make an excel spreadsheet. Simply stated, the product was in the raw sewage state of development

Q: Can they be used on speaker wire?

Unless you're using super thick speaker wire, it should work fine. If zip cord, you may have to split it down the center and apply to one or both wires individually. It may work without splitting the cord, not sure of that.
