Question 1

Does anyone regret not buying a dell u4320q? i want to save $200 but don't want to be kicking myself later on.

I have both DELL as well as this LG monitors, I only use these for professional use (engineering/3D-CAD/etc), and quite frankly both monitors are very good for my use (in fact now I'll never go back to any smaller monitor).  LG has been definitely a better buy due to it being much cheaper.  DELL has better (or should I say more realistic) colors but unless you do some type of work where color matching really matters, you will not be affected.  I do also use this LG monitor with my Adobe suite (mainly illustrator, acrobat, and photoshop) but for my purposes I have not seen any problem whatsoever.  The only complaint about this LG monitor is that it does not charge my LG Gram 2-in-1 laptop even though BOTH are made by the same company LG!  That I find as very annoying because instead of one USB-C cable between the laptop and the monitor, I had to buy a 100W power adapter for powering the laptop. On the other hand, my wife's smaller DELL laptop charges just fine with this monitor so it seems that this monitor cannot handle more then 60Watts even though its manual says it can charge up to 65watts.  Going back to your question, I don't believe you'd be disappointed for buying this over a DELL for professional use. I can't say anything for games/etc as I have not used my DELL or LG monitors for gaming.

Question 2

With 4 youtube videos per quadrant, can you hit the maximize button on each and have it fill up only the respective quadrant not the whole 43" screen?

If you have a video card support more than 4 outputs, you can connect all 4 hdmi as a 2k desktop combine as a one large 4K desktop but with 4 distinct boundary. When hit maximum it will only maximize to each quadrant

Is this monitor really 46 lbs?  Seems like most 43" monitors are all between 30-40 lbs.

Question 4

In 4-split screen mode, does it support multi-stream transport (mst) on the displayport?  so the 4 screens will show up as 4 monitors in windows 10?

Yes it does.  Transfer of data.... pc to pc...  with just the mouse on monitor... drag and drop.
Or cut and paste from one pc on the right side of screen and fill the blanks in the left side on completely different pc that's hook to it.
Love this monitor. 
Would buy two more if it goes back on sale for $299.99 again.
