Will these detect if some one is vaping in the house?

They detect smoke particles, not vapor droplets.

What's the expiration date on these smoke detectors?

The units are rated at 10-year battery. After install of multiple units, 1 unit has issue with the test button. The Fire Prevention Bureau inspection person wasn't keen on units. I'm not noting an expiration date (I would expect 10 yrs from mfg. date).

do these have a light that flashes to see if they are working? Can I use a stick to press the button or do I need to climb on a ladder?

No light that flashes, but you can use a stick to press the button to check, do not need a ladder, my company has bought over 200 and only had a problem with one, these are good quality.
